Description

Podoplanin also termed as Aggrus/gp36/E11 antigen, as is a mucin-type transmembrane glycoprotein with high content of sialic acid, widely expressed in various specialized cells throughout the body. It is a single pass type I membrane proteins. It was first identified in kidney glomerular podocytes epithelial cells and is also known as marker for lymphatic endothelium and alveolar epithelium. The Podoplanin is also expressed in osteocytes and osteoblasrs, mesothelial cells, epidermal basal lateral cells, tooth germ epithelial cell, salivary gland myoepithelium, prostate and many other body cell types (1). The Podoplanin gene promoter region lack the TATA and CAT box but has a CpG island with putative Sp1 and Sp4 sites. Such promoters are usually at the 5'regions of the housekeeping genes ad CpG region hypermethylatioin is linked to transcriptional repression (2). Podoplaniin binds to its cognate receptor C-type lectin like receptor 2 (CLEC-2) which mediate platelet aggregation without blood coagulation and plasma factors (3). Podoplanin has a platelet aggregation stimulating domain (PLAG) of which only the Thr52 is O-glycosylated with disialyl-core 1 is an extra cellular domain. The binding of Podoplanin to CLEC2 is dependent upon the glycosylation state of the PLAG domain. The Podoplanin has three tandem repeats of the PLAG domain which are conserved in Podoplanin homolog of the rat, mouse, dog, human and other species. The physiological function of Podoplanin is still unknown, but may play a role in tissue development and repair as well as in carcinogenesis. The Podoplanin deficient mice die at birth due to respiratory failure. Podoplanin also promotes tumor cell proliferation, migration and invasion. Human Podoplanin gene code for a highly glycosylated protein with162 amino acids (MW 38kDa). The Podoplanin selective antibodies were generated against two peptides taken from human Podoplanin for the extracellular domain close to PLAG domain and form C-terminal domain.
